摘 要

氧化石墨烯(Graphene oxide,GO)因具有独特的理化性能,被广泛用于多种领域。但对其毒性作用以及分子机制了解不完全。因此了解氧化石墨烯的毒性及其致毒机制具有重要意义。miRNAs是一种微小的非编码RNA分子,其在生物体中具有调节各种生理过程的功能。在真核生物中,miRNA是基因表达的重要调节因子,可在转录或转录后水平上进行调节,能调节某些特定细胞和组织类型的靶基因表达。模式动物秀丽线虫的遗传背景目前研究地较为清楚,成长周期短,在毒理实验上具有极好的优越性。实验的前期研究对秀丽线虫mir-235突变体和野生型秀丽线虫(N2)进行氧化石墨烯暴露后,研究其在氧化应激、运动行为的差异,确定mir-235对氧化石墨烯是具有调控作用的。同时确定候选靶基因daf-12在调控氧化石墨烯致秀丽线虫毒性的影响。并在此基础上确定mir-235daf-12的靶向关系,利用qRT-PCR对比分析不同浓度GO暴露以及未暴露的mir-235突变体线虫体内daf-12表达水平的不同以及变化。确定靶向关系后,解析daf-12在调控GO毒性以及介导的信号通路。本实验解析了秀丽线虫mir-235经由靶基因daf-12调控GO转运以及毒性效应的分子机制,同时进一步地解析daf-12调控GO体内转运以及毒性效应是通过什么样的网络。为之后继续在研究秀丽线虫mir-235如何调控氧化石墨烯提供参考。

Abstract

Graphene oxide (GO) is widely used in various fields due to its unique physicochemical properties. However, its toxic effects and molecular mechanisms are not fully clear. Therefore, it is important to study the toxicity of GO and its toxic mechanism. miRNAs are small non coding RNA molecules that have the function of regulating various physiological processes in living organisms. In eukaryotes, miRNAs are important regulators of gene expression that can be regulated at the transcriptional or post-transcriptional level and can regulate the expression of target genes in certain specific cell and tissue types. The genetic background of the C. elegans is relatively clear, so it has excellent advantages in toxicological experiments. In previous study, after exposure to GO in the mir-235 mutant and wild-type C. elegans (N2), the differences endpoints were studied to determine the mir-235 has a regulating effect to GO toxicity. Simultaneously, we determined the effect of candidate target gene daf-12 in regulating GO-induced nematode toxicity. Then the relationship between mir-235 and daf-12 was determined, and qRT-PCR was used to compare the level of daf-12 expression in mir-235 mutant nematodes in exposure to different concentrations of GO. After determining the targeting relationship, daf-12 in the regulation of GO toxicity and mediated signaling pathways are studied. As a result, the molecular mechanism of mir-235 regulating GO toxicity via the target gene daf-12 was analyzed. Our work will provide clues for further study of how mir-235 regulates toxicity of GO.

miRNA调控氧化石墨烯毒性研究进展

miRNAs是一种微小的内源性非编码RNA分子,其在生物体中具有调节各种生物过程的功能。在真核生物中,miRNA是基因表达的重要调节因子,可在转录或转录后水平上进行调节,能调节某些特定细胞和组织类型的靶基因表达。氧化石墨烯因具有独特的理化性能,被用于多种领域,但对其毒性作用以及分子机制了解不完全。本综述阐明了氧化石墨烯的毒性评价和miRNA在其中的调控作用。

氧化石墨烯的毒理评价

纳米材料氧化石墨烯(GO)是具有原子层厚度的石墨烯衍生物,其具有独特物理化学性质,被认为在生物传感器、药物载体及生物成像的研究与应用中具有潜力的工程纳米材料[1.2]

动物实验评价

在对GO的毒性效应的研究中,目前已有的研究是:用人肠癌细胞系MGC803、人乳腺癌细胞系MCF-7、MDA-MB-435、即肝癌细胞系HepG2、HDP、SH-SY5Y细胞A549、小数成纤维细胞系L929等细胞进行的离体GO毒性评价;以及用小鼠、大鼠、斑马鱼、秀丽线虫等模式动物进行的体内毒性试验[3.4.5]

在小鼠的实验中,GO主要积聚在肝脏、肺部、脾脏或肾脏中,并且小鼠的肾脏很难清除积聚的GO[2]。目前人们认为氧化应激、炎症活化和细胞凋亡是证明GO毒性的重要细胞和分子机制。Toll样受体4的诱导可能是GO诱导的巨噬细胞坏死的主要分子机制[6]
